Indirect Fluorescent Antibody test for Diagnosis of Clonorchiasis


Abstract
Indirect immunofluorescent antibody tests were performed with clonorchiasis patient¢¥s sera and rabbit¢¥s sera that were infected with Clonorchis sinensis. Clonorchis antigen was prepared from lyophilized adult worms of C.sinensis by defetting with ethyl-ether before extracting with barbital buffered saline and that reacted
with agarose.
Several rabbits were infected with C.sinnsies per os in a single dose, and antisera were taken in 6 weeks after the administration and 54 clonorchiasis patient¢¥s sera were obtained.
As controls, various other parasitic infected person¢¥s sera and parasite free healthy person¢¥s sera were used.
The results were as follows:
1) The sera from the rabbits to which administered with 100 and 500 metacercariae of C. sinensis showed the titers from 1 : 12,800 to 1 : 51, 200 and from 1 : 25, 600 to 1 : 51, 200 respectively in 6 weeks after the administrating in indirect immunofluorescent antibody tests.
2) Indirect immunofluorescent antibody tests showed positive rates of 87. 0 per cent in the human subjects that were detected of Clonorchis egg, in the case of the titers more than 1 : 40.
3) As controls, the sera were from those infected with other parasites, for example, Paragonimus, Ascaris, hookworm, Hymenolepis, Entameba, Trichocephalus, Taenia and Giardia, showed the titers less than 1 : 20.
